Tight Junction-Proteine bei entzündlichen Darmerkrankungen

Im entzündeten Colon, bei mikroskopischer Colitis und bei Colitis ulcerosa vor und nach Therapie werden Mechanismen der epithelialen Barrierestörung mittels Impedanz-Spektroskopie, Conductance Scanning, konfokaler Mikroskopie sowie Western-blotting und quantitativer RT-PCR analysiert. Dazu werden die epithelialen Tight junction-Moleküle ZO1, Occludin, die wichtigsten Claudine sowie Tricellulin charakterisiert. Daneben werden auch Apoptoseraten sowie epitheliale Läsionen erfasst.

Weiterhin werden Mechanismen der entzündlichen Regulation der epithelialen Tight Junction und der sie aufbauenden Moleküle im HT-29/B6-Zellkulturmodell durch Zytokine sowie durch luminale Faktoren studiert und zwar hinsichttlich Expression, Halblebenszeit, subzellulärer Verteilung, mRNA-Expression samt mRNA-Halblebenszeiten, Translationseffizienz unter der Regulation von miRNAs, Promotoraktivität und intrazellulärer Signaltransduktionswege.
