Barrierefunktion von Tight Junctions in Keratinocyten, Hautäquivalenten und in der Haut
Die Haut ist eine wichtige Barriere des Körpers, die den unkontrollierten Verlust von Wasser und Elektrolyten sowie das Eindringen von Fremdstoffen
verhindert.
Tight Junction (TJ) -Proteine und -Strukturen wurden auch im mehrschichtigen Epithel der Epidermis von Maus und Mensch nachgewiesen. Untersuchungen an Mäusen, denen bestimmte TJ-Proteine in der
Epidermis fehlen, zeigen eine Verminderung der epidermalen Barrierefunktion. An kultivierten humanen und murinen Keratinocyten konnte gezeigt werden, dass sie funktionelle TJs mit deutlichem
transepithelialen Widerstand und eingeschränkter Permeabilität für molekulare Tracer bilden können. Inwieweit diese Ergebnisse auf die menschliche Epidermis bzw. Haut übertragbar sind, welche
Selektivität TJs in Keratinocyten und in der Epidermis aufweisen und welche TJ-Moleküle dabei eine wichtige Rolle spielen, ist noch unklar.
In diesem Projekt soll der Frage nach der Barrierefunktion von keratinocytären TJs für Tracer unterschiedlicher Größe und Ladung und der dafür wichtigen TJ-Proteine nachgegangen werden. Als Modellsysteme werden kultivierte Keratinocyten sowie Hautäquivalente und ex-vivo Hautmodelle gesunder und psoriatischer Haut verwendet. Ein besonderes Augenmerk wird auf die Übertragbarkeit von Ergebnissen aus primären Keratinocytenkulturen auf Hautäquivalente und auf die Haut gelegt.
